Studies on the Determination of Bile Pigments. Iv. Spectrophotometric Determination of Free and Total Bilirubin in Serum.

چکیده
The absorbance at 450 mJ.Lof serum diluted with acidified ethylene glycol is used as a measure of the serum’s total bilirubin content in a single-tube, single-extraction method for rapid determination of free and total bilirubin in 0.1 ml. of serum. With the addition of chloroform, free bilirubin is extracted into the chloroform but conjugated bilirubin remains in the ethylene glycol. Free bilirubin is determined from the absorbance at 450 m in the chloroform layer. Conjugated bilirubin is calculated from the difference. Absorbance readings at 520 compensate for the absorbance at 450 m/L, due to hemoglobin in the serum. Comparison with the Malloy-Evelyn procedure showed equivalent levels of total but higher levels of conjugated bilirubin.

A study of six representative methods of plasma bilirubin analysis.
The large number of modern methods which have been published for the determination of total bile pigments in the blood plasma indicates the difficulties of the estimation and suggests the need for a reliable standard method. In view of the present importance of plasma bilirubin levels in the diagnosis and treatment of jaundiced subjects, six analytical procedures have been studied, each chosen ...
متن کاملCatalytic Spectrophotometric Determination of Traces of Tellurium (IV)
A Simple, rapid and sensitive method has been developed for determination of trances of tellurium (VI) (0.096-1.250 mg/mL) based on its catalytic effect on the reduction reaction of toluidine blue by sulfide ion at pH 4. The reaction is monitored spectrophotometrically by measuring the decrease in absorbance of the dyestuff at 628 nm by the fixed time method. The detection...
متن کاملHepatic accumulation and intracellular binding of conjugated bilirubin.
After the intravenous injection of unconjugated [(3)H]bilirubin into normal Sprague-Dawley and Wistar R rats, radiolabeled bile pigments rapidly accumulated in the liver. By 1.5 min after injection, an average of 36% of the injected isotope was present in liver homogenates. Between 3 and 15 min, 37-64% of the total intrahepatic radiolabeled bilirubin was conjugated, as demonstrated by extractio...
متن کاملStudies on the determination of bile pigments. I. Standard of purity for bilirubin.
HE ACCURACY of the standardization for determination of bilirubin is at present open to question. This is a result of the fact that commercially available bffirubin preparations vary considerably in their chromogenicity in the azobilirubin reaction (1) and because a reliable standard of purity has never been established. Furthermore, difficulty in preparing biirubin solutions has led many worke...
متن کاملFlow Injection Spectrophotometric Determination of Sulphide by Catalytic Reduction of Toluidine Blue
Sulphide was determined spectrophotometrically at 628 nm and pH 6.7 based on its catalytic reduction of toluidine blue in the presence of Se(IV) using a flow injection system. After optimization of the measuring conditions the calibration curve was found linear up to 25 mg/mL and the detection limit was 0.3 mg/mL (30 mL injections). The ...
